※ Paraspeckle introduction Paraspeckles are mammalian-specific nuclear bodies that often lie adjacent to splicing speckles, containing several multifunctional proteins, including PSF (PTB-associated splicing factor), p54nrb, PSP1andRBM14/CoAA/PSP2 (here- after referred to CoAA) (1), the long noncoding RNA NEAT1 assembles these protein components and is critical for paraspeckle formation. DIS3 was proved to be involved in controlling the formation of paraspeckles, nuclear bodies that are organized around NEAT1 lncRNA, whose short form was overexpressed in cells with mutated DIS3 (2). Paraspeckles regulate gene expression through a number of mechanisms, including sequestration of specific transcription factors and/or hyperedited mRNAs (3). Besides, RNA editing by the enzyme ADAR (Adenosine deaminase acting on RNA) was proved to occur in paraspeckles (4).
